Research into their pharmacological properties has shown that cannabinoids are not all created equally, however: some matter more than others, from a manufacturing process point-of-view. Four key gateway cannabinoids hold the door through which all others must pass:  CBGA, CBNA, CBGV, and CBGVA.  Following the most prevalent synthesis pathway in the plant, CBGA is formed from precursors; downstream enzymes then determine if CBGA diversifies to become tetrahydrocannabinolic acid (THCA), cannabidiolic acid (CBDA), cannabichromene acid (CBCA), or cannabigerol (CBG). Then, further reactions by synthases give rise to a multitude of the most common cannabinoids that so richly endow the plant, including THC and CBD.

InMed’s progress brings scale-up and commercialization of its disruptive biosynthesis process closer to fruition. Traditionally, sourcing cannabinoids has been accomplished by growing, harvesting, processing and purifying (via extraction) the cannabis plant. The extraction process can be expensive, and can result in unwanted by-products and impurities. Biosynthesis mimics the cannabinoid-creation process found in plants by utilizing a microbial host, resulting in pure, individual cannabinoids that will be devoid of the by-products and impurities seen with extraction. The InMed approach is to introduce cannabinoid DNA into E. coli bacteria that results in a biofermentation process, enabling a laboratory-based process under tightly controlled conditions. The process can be tailored to produce any of the 90+ “downstream” cannabinoids found naturally in the cannabis plant. InMed’s biosynthesis program will provide an alternative to the agricultural approach in a low cost and high quality process.  Currently, the Company is actively employing this production chassis to biosynthesize compounds for its pharmaceutical research programs. InMed currently has two lead candidates in its drug pipeline. The condition, which currently has no approved treatment or cure, has been called “The Worst Disease You’ve Never Heard Of” by the Dystrophic Epidermolysis Bullosa Research Association of America. INM-750 works two ways. First, it addresses the primary hallmarks of the disease: wound-healing, pain, inflammation, itch, and infection. Second, this compound may have the potential to reverse the course of the disease itself in some patients by upregulating compensatory keratins in the skin to replace defective ones.

The second candidate is INM-085 for the treatment of glaucoma, a leading cause of blindness, according to the Glaucoma Research Foundation. The drug reduces the elevated intra-ocular pressure that is often associated with glaucoma. It is targeting a large market. The National Institutes of Health estimates that more than 3 million Americans currently have glaucoma.

The CSE25 Index includes the top 25 securities contained within the CSE Composite Index based on market capitalization. If you have any doubt that InMed Pharmaceuticals, Inc. (CSE: IN) (OTCQB: IMLFF) is on the right track as it moves to protect its biosynthesis technology for cannabinoids, then a word from a famed figure may make you change your mind. Known worldwide for his investing prowess, Warren Buffett has said, “I don’t want a business that’s easy for competitors. I want a business with a moat around it with a very valuable castle in the middle” (http://dtn.fm/9al5F). Naturally, the Sage of Omaha was referring to an economic moat and not its tangible water-filled counterpart. One obvious way of constructing such an economic moat would be by registering a patent, which is exactly what InMed is planning to do. The company, which specializes in the research and development (R&D) of novel, cannabinoid-based drug therapies, has announced the filing of a provisional patent application for its proprietary biosynthesis process for the manufacture of cannabinoids that are identical to those found in nature (http://dtn.fm/I8g3M).

This filing is the first step in setting up defenses. Under U.S. patent law, a provisional patent application filed with the United States Patent and Trademark Office (USPTO) allows the filer to get a filing date and book a place in the queue. But, just as importantly, it affords protection for up to 12 months without filing a complete patent application. The provisional application may later be converted into a non-provisional one or be replaced by a regular patent application. The process in Canada, although not the same, is very similar.

To obtain protection outside North America, the Patent Cooperation Treaty (PCT) offers one streamlined route. The PCT procedure facilitates a patent application in all contracting states that are signatories with one filing, obviously a boon, since 145 sovereign states are members of the PCT. The PCT process allows the filing of an “international patent application,” which acts as an initial application for a patent in any member country. However, an “international patent application” never matures into an “international patent”. No such animal exists. A PCT filing simply reduces the burden of individual filings to contracting states and acts as a patent application to the contracting state in which the patent is meant to go into effect. Patents are issued by sovereign states and are always limited to national boundaries.

InMed’s provisional patent application will protect the biosynthesis process it has developed for the production of cannabinoids. In itself, biosynthetic production of chemicals is nothing new, but it really took off in 1978 after Genentech succeeded in modifying Escherichia-coli (E-coli) bacteria to produce synthetic insulin, a breakthrough credited with saving thousands of lives. Insulin is required by diabetics, whose bodies either do not produce the hormone or handle it inefficiently, in order to absorb glucose. In its absence, blood sugar increases to levels that may damage internal organs. Before biosynthesis, insulin was produced from the pancreas of pigs and cows in a costly, rather inhumane process.

InMed’s biosynthesis technology involves a process that can be applied to manufacture all the naturally occurring cannabinoids, through which the DNA of the cannabinoid to be produced is applied to Escherichia-coli (E-coli) bacteria. The E-coli DNA is then removed, leaving just the cannabinoid DNA, which can then replicate and multiply. This process combines the inherent safety and known efficacy of the natural drug structures with the convenience, control and quality of a laboratory-based manufacturing process.

InMed’s biosynthesis methodology will likely be used to produce the less naturally occurring phytocannabinoids such as delta-8 tetrahydrocannabinol (Δ8 THC), cannabidivarin (CBDv), and tetrahydrocannabivarin (THCv), although, for the present, the company is not saying. Regardless of which cannabinoid castles InMed decides to build, they will all be protected by a sturdy economic moat.

The Vancouver, Canada-based company continues its research and development (R&D) into the therapeutic benefits of cannabinoids, an activity that holds the promise of exceedingly rich reward. Peer valuations have skyrocketed, increasing the likelihood that unicorns may soon appear on the cannabis biotech landscape. With a triad of valuable core assets, it won’t be surprising if, sometime in the future, InMed turns out to be one of them.

InMed Pharmaceuticals has more to offer than the typical drug discovery and development company. Its triple play comprises a proprietary technology based on biosynthesis, a bioinformatics system, and a robust drug development pipeline, any one of which may propel this innovative biotech to success. The company has developed a process to manufacture cannabinoids by applying cannabinoid DNA to Escherichia-coli (E. coli) bacteria. The E. coli DNA is then removed, leaving just the cannabinoid DNA, which can then replicate in a process known as biosynthesis. Biosynthesis is widely employed in the pharmaceutical industry and is used, for example, to manufacture the insulin upon which millions of diabetics rely.

InMed’s bioinformatics system, employing proprietary algorithms, identifies the cannabinoids most likely to have beneficial therapeutic effect in serious diseases with high unmet medical needs. The program integrates data from numerous bioinformatics databases, including a database on the structure of currently approved pharmaceutical products and an extensive proprietary database of over 90 individual cannabinoid drugs found in cannabis. This bioinformatics tool has already identified several therapies, including the company’s two lead pipeline candidates, INM-750 and INM-085.

INM-750 is for the treatment of a rare genetic connective tissue disorder called epidermolysis bullosa (EB). EB is an orphan pediatric disease characterized by extremely fragile skin that affects roughly one out of every 20,000 births in the United States. The condition, which currently has no approved treatment or cure, has been called “The Worst Disease You’ve Never Heard Of” by the Dystrophic Epidermolysis Bullosa Research Association of America. INM-750 works by replacing missing keratins in the skin with specially selected cannabinoids in an effort to modulate the painful manifestations of EB. It has potential global market revenues of approximately $1 billion.

INM-085 targets glaucoma, a leading cause of blindness, according to the Glaucoma Research Foundation. The drug reduces the elevated intra-ocular pressure that is often associated with glaucoma and so holds the promise of providing relief to the more than three million Americans who currently have glaucoma.

Both INM-750 and INM-085 are in the pre-clinical phase, and InMed is taking the lead with the former, which it is hoping to advance to phase I clinical trials in 2018. If all goes well, INM-750 is expected to be in phase III by 2020, setting InMed on a trajectory, if peer comparisons are any indication, to a much higher valuation than its current one of under $50 million. Scioderm, for example, was acquired by Amicus two years ago for $847 million. This acquisition was based on the results of a phase II study of its sole clinical asset Zorblisa, which involved only 42 patients. Zorblisa, for the treatment of EB, is currently in phase III.

InMed’s pipeline is in good hands. Its chief scientific officer is Dr. Sazzad Hossein, who brings more than 20 years of academic and industrial experience in new drug discovery and natural health product development to the C-suite. He was, at one time, group leader and senior scientist at Biotechnology Research Institute of National Research Council Canada, the Government of Canada’s prime biotechnology research organization, where he set up a pharmacology laboratory to evaluate the safety and efficacy of new drugs under development in the areas of cancer and cardiovascular and ocular diseases. He is also the author of more than 40 peer-reviewed papers, primarily in the pharmacology, genetics and nutritional sciences.

InMed’s chief medical officer is Dr. Ado Muhammed. Muhammed joined the company from GW Pharmaceuticals, developer of Sativex, where he was associate medical director, distinguishing InMed as the only biopharmaceutical company in North America with a senior GW team member on its executive team.

InMed Pharmaceuticals, Inc. (CSE: IN) (OTCQB: IMLFF), a biopharmaceutical company focused on identifying and commercializing prescription drugs using non-THC cannabinoids, recently reached a research and development agreement with tissue engineering company ATERA SAS of France (http://dtn.fm/Yv6Bd). The fundamental benefits of this agreement for InMed, as it eyes a clinical trial program, are a proprietary assessment tool, a biosynthesis manufacturing process, and drug development programs.

Vancouver-based InMed’s lead compound is INM-750, a proprietary topical cannabinoid product candidate aimed at treating skin disease epidermolysis bullosa (EB). EB is a skin disease which retards wound healing and can cause pain, itching, and inflammation. INM-750 may reverse the disease, for which there are currently no approved therapies. In its corporate presentation, InMed said that it sees a potential global market of $1 billion for INM-750, if approved.

ATERA SAS, as part of the agreement, will develop 3D human skin models that will assist InMed in assessing INM-750’s effectiveness in treating EB in vitro ahead of upcoming clinical trials estimated by InMed to take place in 2018, 2019 and 2020. ATERA SAS will produce 3D skin models in a controlled environment, creating a unique tool for drug development screening. ATERA SAS is expected to develop the 3D models from EB patient biopsies in order to help InMed assess INM-750.

ATERA SAS is a tissue engineering company tasked in this agreement with developing advanced human tissue models to assist InMed in analyzing pre-trial tests. The French company will also investigate INM-750 at the ultra-structural cellular and molecular levels. It will create 3D in vitro reconstructed human full thickness skin models of both normal and EB-derived skin cells.

At InMed, Dr. Ado Muhammed, MD, DPM and MFPM, is the chief medical officer. He previously served as associate medical director at GW Pharmaceuticals, where he was key in leading the company in the development and FDA approval of one of the first cannabis drugs. InMed is the only cannabis biotech company led by a former GW Pharmaceuticals executive.

The results are that THC activates the CB1 receptor to provide analgesic relief without side effects. The study results suggest that THC could provide a novel approach to offer that relief through the peripheral application of cannabinoids.

The article is titled, “Delta-9-tetrahydrocannabinol decreases masticatory muscle sensitization in female rats through peripheral cannabinoid receptor activation.” The study was co-funded by Canadian non-profit organization Mitacs and InMed Pharmaceuticals. In a news release, Hossain, co-author of the findings report, said, “This study sets the stage for advanced work in various pain models to explore the role of several cannabinoid compounds, applied as topical agents, to target the CB1 and other pain-related receptors.”

The result could be a valuable approach in the cannabinoid treatment of severe pain. The model used in this study mimics the muscle pain of temporomandibular joint disorders (TMD) that affect the jaw muscle. TMD is a chronic pain condition that’s difficult to treat.

Moving toward offering the very first therapy for treating epidermolysis bullosa (EB), InMed Pharmaceuticals, Inc. (CSE: IN) (OTCQB: IMLFF) recently announced it has signed an agreement with Pharmaseed Ltd. (http://dtn.fm/o5XNU) to develop a final formulation for INM-750, InMed’s lead compound, which is being developed as a therapy for EB and for other potential dermatological and wound-healing applications.

EB is a group of inherited connective tissue diseases sharing a common manifestation of very fragile skin that is prone to easily blistering or tearing. An orphan disease that currently has no approved treatments and has significant unmet medical need, EB results from a defect in anchoring between the dermis and the epidermis and is most commonly caused by the absence of specific keratins in the skin. In addition to its effects on the skin, EB can also affect bodily systems and internal organs.

InMed’s INM-750 is a proprietary topical product candidate that includes multiple cannabinoids as its active ingredients, which were chosen to modulate keratin levels in EB patients in anticipation of having a disease-modulating effect. A secondary reason for selecting these cannabinoids was to address EB symptoms, including inflammation, wound healing, skin regeneration, itching and pain. Through the agreement with Pharmaseed, which is the largest GLP-certified preclinical contract research organization in Israel, InMed is taking another step toward moving INM-750 to its first clinical trial. Under the terms of the agreement, Pharmaseed will develop a final formulation for INM-750 for ongoing R&D, including IND-enabling pharmacology and toxicology studies and eventual clinical studies. The contract also includes the development of assay methods for manufacturing, stability, quality assurance and other methods of analysis.

InMed Pharmaceuticals, Inc. (IMLFF) is a Player in the Billion Dollar Cannabis Biotech Market

Despite the fire-breathing from Attorney-General Jefferson Beauregard Sessions III, there is a distinct feeling that marijuana has entered the mainstream now that Goldman Sachs has initiated coverage on GW Pharma (NASDAQ: GWPH). GW Pharma is the biopharmaceutical that developed the cannabinoid-derived Sativex to treat spasticity due to multiple sclerosis (MS), now approved by regulatory authorities in 29 countries. The success of GW Pharma and other pharmaceutical companies focused on cannabinoid research has produced an exciting new field with a lot of promise, cannabis biotech, and InMed Pharmaceuticals, Inc. (CSE: IN) (OTCQB: IMLFF) is staking a position in that emerging market. The Vancouver, Canada-based outfit is a pre-clinical stage biopharmaceutical company that is currently testing cannabinoid derivatives to treat epidermolysis bullosa and glaucoma and a full pipeline of other drug candidates.

Cannabinoids are turning out to be big business. GW Pharma presently has a market cap of around $3.0 billion, yet Goldman Sachs, in that initial report, intimated the company might be worth more. It set a target price of $189 on the stock, currently trading at just above $116, which would value the company at close to $5 billion. GW Pharma is set to file a New Drug Application (NDA) with the FDA in the first half of 2017 for Epidiolex, the company’s cannabis-based treatment for rarer forms of epilepsy.

Axim Biotechnologies, Inc. (OTCQB: AXIM) is another player showing value. The company owns the patent for chewing gum as a delivery method for cannabinoids, which it has developed into two separate products. Its current market cap is about $500 million. Cara Therapeutics, Inc. (NASDAQ: CARA), meanwhile, is researching therapies that target the CB1 and CB2 cannabinoid (CB) receptors. Its most advanced CB compound, CR701, is in preclinical development. The company’s market value is about $556 million. Zynerba Pharmaceuticals, Inc. (NASDAQ: ZYNE) has a different focus. This biotech is studying synthetic cannabinoid therapeutics across multiple therapeutic areas. It has a market cap of over $220 million.

Research into the properties of cannabis was galvanized after Raphael Mechoulam and Yehiel Gaoni first isolated delta-9-tetrahydrocannabinol (THC) in 1963. Then, cannabinoid research became an even more exciting area of scholarship in 1992 when the first endocannabinoid, anandamide, was discovered. An endocannabinoid is a cannabinoid that is manufactured by the body. The fact that our bodies were producing the very same substances found in cannabis raised some very interesting questions.

Now we know that residing within most life forms (basically everything on our planet with the exception of insects) there is an extensive molecular signaling network known as the endocannabinoid system, which controls many functions. It has been said that ‘the endocannabinoid system is possibly the single-most important system within our entire bodies – responsible for maintaining homeostasis’ (http://dtn.fm/8Q5dt). Consequently, the aim of much cannabinoid research has been to provide cannabinoids to the body when, for one reason or another, the body fails to provide adequate quantities.

Presently, InMed Pharmaceuticals has two promising drug candidates undergoing pre-clinical trials. The first, INM-750, is for the treatment of a rare genetic connective tissue disorder called epidermolysis bullosa (EB), which affects roughly one out of every 20,000 births in the United States. There is an estimated patient population of about 25,000 with this condition in the U.S. alone. Globally, the market potential is about $1.0 billion.

The second drug, INM-085, treats glaucoma, a leading cause of blindness. INM-085 works by reducing the elevated intra-ocular pressure that is often associated with glaucoma. The National Institutes of Health estimates that more than three million Americans currently have glaucoma, and that the disease has blinded more than 120,000. INM-085 has a global market potential of close to $6.0 billion. Cannabinoid research is proving to be a truly exciting field.

InMed Pharmaceuticals, Inc. (IMLFF) Gains Attention for Its Operational Developments and Leadership Team

InMed Pharmaceuticals, Inc. This tool has helped the company identify two therapies, one for epidermolysis bullosa (EB), a rare skin disease found in children, and one for glaucoma, a degenerative condition that causes damage to a person’s optic nerve. In addition to all of the above, the company has now developed a method of biosynthesis similar to that used to create synthetic insulin, but for cannabinoids. This process is not only cheaper, but quicker, and it gives access to cannabinoids that are not currently profitable to turn into drugs.
